A groundbreaking study led by Josh Conway has found that a combination of three drugs may help target cells linked to aging and cancer. The treatment combines dichloroacetate, metformin, and a low dose of navitoclax. Researchers found that the combination helped extend the lives of older mice while leaving healthy cells unharmed.
The Dual Threat of Aging and Cancer
As we get older, our bodies build up senescent cells, which are damaged cells that stop dividing but do not completely disappear. Instead, they can release harmful substances that cause inflammation and damage nearby tissues. Over time, this damage may contribute to age-related health problems and may also play a role in cancer development.
These cells can be especially difficult to deal with because they are no longer functioning normally but remain active in the body.
Cancer treatments such as chemotherapy are designed to kill or slow cancer cells. However, some treatments can also increase the number of senescent cells. This has led researchers to search for ways to remove these damaged cells while protecting healthy ones.
One of the drugs in the study, navitoclax, has shown potential for targeting these cells. However, higher doses can cause serious side effects, including thrombocytopenia, a condition in which the body has too few platelets.
A New Three-Drug Approach
To address these challenges, researchers combined dichloroacetate and metformin with a low dose of navitoclax. They called the combination DMA.
The three drugs work together by targeting the way damaged and cancerous cells produce energy. This gives researchers a different approach to dealing with cells that may contribute to aging and cancer.
In experiments, the DMA combination performed better than any of the individual drugs used alone. It killed about 60% of damaged and senescent cells, with even stronger results under conditions designed to more closely resemble the body’s natural environment.
Importantly, healthy cells were not harmed in the experiments.
What Could This Mean for Healthy Aging?
The most promising part of the study is its potential connection to healthier aging and longer life. Researchers found that the drug combination helped extend the lives of older mice by targeting harmful cells that build up with age.
This could eventually lead to new approaches for delaying some age-related diseases and improving health span, or the number of years a person remains healthy.
However, the findings are still early. The research has been conducted in cells and mice, so it does not yet show that the treatment can extend human lifespan.
What Could This Mean for Cancer?
The same approach may also have potential in cancer research. Because the DMA combination targets both senescent and cancerous cells, researchers are exploring whether it could offer another way to deal with cells involved in tumor development.
However, this does not mean DMA is currently a cancer treatment. More research is needed to understand how effective and safe the combination would be in people.
Navitoclax is also not approved for human use, which means the treatment is still far from being available as a standard therapy.
The Bigger Picture
The study offers an interesting new direction for healthy aging research, while also showing possible applications in cancer research. By targeting damaged cells that accumulate with age, researchers may be able to find new ways to support healthier aging and potentially reduce some age-related health risks.
For now, DMA remains an experimental treatment. The results are encouraging, but further studies are needed before researchers can determine whether the same benefits and safety can be seen in humans.
